LRIG1 is a gatekeeper to exit from quiescence in adult neural stem cells

چکیده

Abstract Adult neural stem cells (NSCs) must tightly regulate quiescence and proliferation. Single-cell analysis has suggested a continuum of cell states as NSCs exit quiescence. Here we capture characterize in vitro primed quiescent identify LRIG1 an important regulator. We show that BMP-4 signaling induces dormant non-cycling state (d-qNSCs), whereas combined BMP-4/FGF-2 distinct poised for cycle re-entry. Primed (p-qNSCs) are defined by high levels CD9, well interferon response signature, can efficiently engraft into the adult subventricular zone (SVZ) niche. Nutrition-Responsive Glia Control Exit of Neural Stem Cells from Quiescence

The systemic regulation of stem cells ensures that they meet the needs of the organism during growth and in response to injury. A key point of regulation is the decision between quiescence and proliferation. During development, Drosophila neural stem cells (neuroblasts) transit through a period of quiescence separating distinct embryonic and postembryonic phases of proliferation. It is known th...

CDK6 Levels Regulate Quiescence Exit in Human Hematopoietic Stem Cells

Regulated blood production is achieved through the hierarchical organization of dormant hematopoietic stem cell (HSC) subsets that differ in self-renewal potential and division frequency, with long-term (LT)-HSCs dividing the least. The molecular mechanisms underlying this variability in HSC division kinetics are unknown. We report here that quiescence exit kinetics are differentially regulated...

Single-cell expression profiling of human epidermal stem and transit-amplifying cells: Lrig1 is a regulator of stem cell quiescence.

Considerable progress has been made in characterizing epidermal stem cells by microarray analysis of FACS-selected populations. One limitation of this approach is that the gene expression profiles represent the average of the cell population, potentially masking cellular heterogeneity of functional significance. To overcome this problem, we have performed single-cell expression profiling. We ha...

Isolation and Differentiation of Neural Stem/Progenitor Cells From Subventricular Zone of One Adult Rat

Introduction: In adult mammalian brain, neural stem cells are isolated from both the dentate gyrus and subventricular zone. This study aimed to isolate neural stem cells from adult rat subventricular zone and differentiate them into neurons and astrocytes.  Methods: In this study, the whole brain was removed after full anesthesia and creating cervical dislocation. Under a microscope, subv...
